The Keith Porter Imaging Facility in room 079 of the Biological Sciences building contains the following instrumentation:

ZEISS LSM 910 confocal

The ZEISS LSM 910 is a compact confocal laser scanning microscope designed for innovative imaging and smart analysis. This system features a motorized stage for tiling and multi-position time-lapse experiments, combining conventional confocal microscopy with optional high-speed volumetric acquisition capabilities. The LSM 910 offers AI-assisted features including the AI Sample Finder wizard that automatically detects and helps navigate around samples on your slides, streamlining the imaging workflow. LSM Plus processing enhances signal-to-noise ratios and improves resolution through advanced deconvolution algorithms without requiring additional hardware.

The Leica M205 is a bright-field and fluorescence stereomicroscope with motorized controls and magnification of 8-320x. It is equipped with a 5 MP Amscope CMOS color digital camera. Filters include DAPI, GFP and DsRed.

 

 

Zeiss Axio Observer 7 with Apotome .2Zeiss Axio Observer

The Zeiss Axio Observer is an inverted microscope with transmitted and fluorescence illumination. This unit has a motorized Z-axis for optical sectioning. The Apotome attachment allows for structured illumination to eliminate out of focus/scatted light. Filters include DAPI, GFP, and TexasRed. Equipped with both a 5 MP color camera for histology and 6 MP monochrome camera for fluorescence microscopy. More information on the Zeiss Axio Observer configuration and specifications.

 

Leica SP5 confocalLeica SP5 confocal

The Leica SP5 confocal inverted microscope includes a motorized stage for tiling and capture of multiple data points during one time-lapse experiment. The instrument is equipped with ISS fluorescence correlation spectroscopy (FCS) hardware that includes two avalanche photodiode detectors (APDs). Hitachi HT7800 TEMHitachi HT7800 TEM

The Hitachi HT7800 is 120 kV TEM is equipped with an AMT Nanosprint15 B camera, brightfield STEM and an Oxford Xplore EDS detector. Navigation is performed with the high speed CMOS camera and images are taken with the high-sensitivity CMOS AMT Nanosprint15 B camera. Magnification goes up tp 600,000x with a point resolution of 0.20 nm. More information on the Hitachi HT7800 configuration and specifications.

 

Sample Preparation Equipment

  • The PELCO easiGlow Glow Discharge Cleaning System for cleaning TEM grids and hydrophilization of TEM carbon support films, which have the tendency to be hydrophobic. A glow discharge treatment with air will make a carbon film surface negatively charged (hydrophilic) which allows aqueous solutions to spread easily.
  • EMS 3000 critical point drier has a horizontal pressure chamber measuring 30.1 mm internal diameter x 82 mm in length. The chamber has an external water jacket temperature control and specimens are introduced via a removable rear door. The front of the chamber is fitted with a 25 mm diameter window which permits easy viewing of the liquid level.
